Correlation of Human Telomerase Reverse Transcriptase Promoter (hTERT) Gene Methylation and Ageing

Promoter methylation of the hTERT gene in blood DNA has been proposed to be an epigenetic molecular clock because it is negatively correlated with ageing. Saliva is an alternative source of DNA because it contains buccal cells. Currently, the correlation of hTERT promoter methylation and ageing using saliva is not known. This study aimed to determine: first, the correlation between hTERT promoter methylation and ageing in saliva, as a source of non-invasive DNA sampling, instead of blood was determined. Second, the influence of sex on the methylation of the hTERT promoter in ageing was evaluated. A cross-sectional study design was used, and 119 subjects were recruited, consisting of 25 children (1-5 y.o), 42 teens (17-19 y.o), 16 adults (20-50 y.o), and 36 elderly (60 to 84 y.o). Promoter methylation of the hTERT of extracted DNA was determined using the MSRE (methyl-specific restriction enzyme) method. The relationship between age and the percentage of hTERT methylation was assessed using the Pearson test. The percentage of hTERT methylation in saliva DNA was negatively correlated with age r= -0.4305 (p-value <0.05). Negative correlation was also found in men (r= -0.376) and women (r=-0.43). Negative correlation between hTERT and ageing has been confirmed in saliva as a noninvasive sampling method. The benefit of this research in ethical and social considerations may encourage a greater participation in ageing research, particularly in underprivileged communities, thus democratising access to scientific advancements in longevity studies.

Abstrak:
Personal data dissemination is a new type of crime that has emerged in the digital world. The rapid development of technology has led to various innovations in data storage, one of which is through barcode technology. Although it provides convenience, this technology also opens up new gaps in cybercrime, especially the misuse and dissemination of personal data stored in barcodes. The impacts of the spread of personal data include damage to personal reputation due to the widespread dissemination of personal data on the internet, the emergence of cyber bullying and sexual harassment, personal data that has the potential to be sold, and the distress and psychological trauma experienced by victims of personal data dissemination. The purpose of this study is to analyze how law enforcement against perpetrators of abuse of the spread of personal data stored in barcodes is reviewed from the ITE Law and the PDP Law, and identify preventive efforts that can be made to prevent these crimes. This research uses a normative juridical approach method used to examine applicable legal norms related to personal data protection. With a statute approach, books, scientific journals, and the internet. The results showed that, although the ITE Law does not specifically regulate barcode technology, the protection of personal data stored in barcodes is still subject to the provisions of the ITE Law and Law Number 27 of 2022 concerning Personal Data Protection.

With technological advancements, digitalization in business has become a crucial factor in enhancing sales effectiveness, especially for micro, small, and medium enterprises (MSMEs). Toko Sakinah, which has been operating for nearly 35 years in Bengkalis, still relies on conventional sales methods, such as WhatsApp transactions or direct store visits. This approach limits market reach and operational efficiency. Therefore, this study aims to design and develop a web-based digital sales system using the Design Sprint method to improve sales effectiveness at Toko Sakinah.The Design Sprint method is applied to develop the system quickly and systematically, focusing on understanding user needs, prototyping, and testing solutions before full implementation. The result of this research is a sales website that facilitates online product ordering, sales reporting, and interaction between buyers and store owners. This website is expected to expand market reach, enhance sales efficiency, and serve as a development model that can be replicated by other MSMEs.

Abstrak:
The development of social media has transformed the way Generation Z constructs and presents their self-identity. One emerging phenomenon is the use of more than one Instagram account by a single user, known as the first account and second account. This study aims to analyze the use of multiple Instagram accounts by Generation Z in shaping self-image, using Erving Goffman's dramaturgical theory. This research adopts a descriptive qualitative method with a phenomenological approach. The informants consisted of eight 11th-grade students from SMK Negeri Nusawungu who actively used multiple Instagram accounts. Data were collected through observation, semi-structured interviews, and documentation, and analyzed using Miles and Huberman’s interactive model. The findings show that the first account functions as the front stage, where informants present an ideal image with curated content and carefully managed impressions. In contrast, the second account serves as the back stage, used for freer, more spontaneous, and emotional self-expression, with a more selective and private audience. The seven key dramaturgical concepts—mystification, setting, front personal, role distance, stigma, impression management, and frame analysis—were identified in the online behavior of the informants. This study recommends the importance of digital literacy and social empathy in understanding the dynamics of youth identity in digital spaces.

Abstrak:
Abstract, Learning media is very useful, especially in improving students' understanding of concepts, strengthening their retention, and fostering their interest in learning. This study uses a descriptive qualitative approach to understand the problem of limited social studies learning media in grade 3 of SDN Sungai Miai 4 and find relevant solutions. Nusantara Kartu is an innovation in learning media designed to introduce and foster students' appreciation of the diversity of Indonesian culture. Nusantara Kartu media has a physical form like a medium-sized card, printed in color, and equipped with pictures and brief information about cultural elements in Indonesia. Nusantara Kartu media is able to overcome the problem of limited textbooks and student boredom in social studies learning. This media not only creates a fun and meaningful learning atmosphere, but also increases students' active participation and understanding of the diversity of Indonesian culture. Teachers can start trying to create similar media that suits the needs of each class.

Abstrak:
This study investigates the impact of product quality, price, and brand image on the purchasing decisions of Wardah facial moisturizer among female students at the Faculty of Economics and Business, University of Lampung. Wardah was selected due to its popularity among young consumers and its reputation as a halal-certified brand offering affordable, high-quality products. Using a quantitative survey method, data were collected from 115 respondents who had purchased the product at least three times. The analysis, conducted through multiple linear regression, reveals that product quality, price, and brand image significantly influence purchase decisions both individually and simultaneously. These results highlight the importance of these factors in consumer decision-making and offer practical insights for cosmetic companies, especially Wardah, in crafting marketing strategies tailored to the preferences of young consumers. Additionally, the findings contribute to the growing body of marketing literature on cosmetic products in the Indonesian context.

Abstrak:
Garment companies are faced with the challenge of employee turnover intention that has not been fully resolved. This study aims to examine the relationship between job insecurity and workload on employee turnover intention through the mediation role of work stress in the garment industry Mas Arya 2 Semarang. Questionnaires were collected from 96 samples of employee staff using proportional sampling techniques with the criteria of permanent employees in the production department who have worked for at least 6 months. Data analysis used partial least squares (SEM-PLS). The findings of the study explain that job insecurity and workload directly affect employee work stress and employee turnover intention. Work stress indirectly acts as a mediator that influences the relationship between job insecurity and workload on employee turnover intention.

This study investigates the complexity of contextual meaning in political texts posted by public figures on the social media platform X (formerly Twitter). Grounded in Halliday and Hasan’s theory of situational context and Searle’s speech act theory, the research analyzes how political utterances function through locutionary, illocutionary, and perlocutionary acts. The data, drawn from tweets during the 2024 Indonesian presidential election period, were collected using documentation and structured observation methods. The findings indicate that political texts on X are highly layered, combining explicit facts with implicit criticism, ideological stance, and persuasive strategies. Through representative, expressive, and directive speech acts, public figures construct meaning that influences public perception, evokes emotional responses, and fosters political alignment. This study contributes to the understanding of digital political discourse by highlighting how pragmatic strategies shape the interpretation of meaning in online political communication.

Abstrak:
Micro, Small, and Medium Enterprises (MSMEs) are the backbone of the Indonesian economy, but in digital business practices, they often face legal challenges in business contracts, whether with suppliers, distributors, or marketplace platforms. This article examines aspects of legal protection for MSMEs in digital business contracts and identifies inequalities that occur in business relationships with large companies. This study uses a normative-juridical research method by analyzing business contract regulations in Indonesia as well as comparative studies with legal systems in other countries. This study concludes that MSMEs need more proactive regulations in ensuring fairness in digital business contracts so as not to experience exploitation from more dominant parties.

In today's global era, interest in learning has shown a significant decline as many individuals are more focused on social media platforms. Social media is not only popular at the local level but has also become an essential element worldwide in supporting daily activities. Although technological advancements have made long-distance communication with loved ones easier, children today tend to use electronic devices more for completing school assignments than for truly understanding the learning materials. They often perceive learning activities as uninteresting, even though without the learning process, one cannot gain extensive knowledge. The decline in learning interest has become more apparent due to technological advancements that provide fast access to information, making people feel less inclined to read or understand step-by-step processes. As a result, many have abandoned traditional learning methods and prefer to use technology as an educational tool rather than reading books. When someone chooses to read, the materials they select are usually not textbooks but rather novels or other works of fiction that are more engaging and entertaining, as they offer various emotional experiences such as joy, sorrow, and pain. This contrasts with textbooks, which are often perceived as boring and less appealing, leading to a reluctance to read them. Consequently, many individuals prefer using technology to solve problems, as it is considered more practical than reading books.
